Colin held the office of Vice-Regent and Lieutenant of the Kingdom in May 1511. He succeeded to the title of 4th Lord Campbell on 9 September 1513. He succeeded to the title of 3rd Earl of Argyll on 9 September 1513. He succeeded to the title of 3rd Lord Lorne on 9 September 1513. He held the office of High Justiciar [Scotland] in July 1514. He held the office of Lieutenant of the Isles in 1516. In 1528 he obtained the barony of Abernethy, part of the forfeited estate of Archibald Douglas, 5th Earl of Angus. He held the office of Master of the King's Household in 1528. He held the office of Lieutenant of the Borders and Warden of the Marches in July 1528.
According to Gibbs, "he was a prominent statesman both during the minority of [King] James V, when he exerted himself to procure the tranquility of the Isles, and throughout the reign of that King."
